Abstract     Seeds of opium poppy Papaver bracteatum were surface sterilized with both solution of Na-hypoclorite or H2O2 and germinated on agar sodified medium with or without Knop's (1865) salt solution for embryo rapid growth. Callus was obtained from seedlings or epicotyls placed on agar sodified Murashige-Skoog (1962) basal medium. Combinations of growth regulators aded to the medium were examined. Some methods to decrese browing of the cultures, especially adding ascorbic acid to the medium, were also observed. Meristemoids, spheres of closly packed cells, developed on surface of calus cultures, when cultured on basal medium containing 0,1 mg/l 2,4-D in 0,1 mg/l KIN. By remowing growth regulators from the medium, green buds and shoots were formed at high frequency and differetiation of into somatic embryos was also accomplished. Thebaine in ammounts 0,1-0,3% was detected by HPTCL and TCL in callus cultures. The presence of other alkaloids morphin, codein and papaverin were also confirmed, while no narcotin was found.
