Abstract     The effect of sucrose, jasmonic acid (JA) and the darkness on bulb formation of garlic Allium sativum L. cv. Ptujski jesenski was studied in vitro. For shoot induction on garlic basal plates we used Gamborg's B5 medium supplemented with 3% sucrose, 5 microM JA and 5 microM 2-isopentenil adenine (2iP). For bulb induction, explants with developed shoots were transferred onto media with 3%, 8% and 12% sucrose. Parrallely, we studied the effect of darkness on bulb formation on media supplemented with 3% and 8% sucrose in presence of 5 microM JA. 8% sucrose significantly increased the percentage of shoots which formed bulbs, the diameter of bulbs and the number of bulbs per one basal plate. The effect was pronounced on the medium supplemented with JA. Growth of explants in the darkness proved to be inefficient in stimulation of bulb formation. We also studied different treatments of transferring microbulbs into in vivo conditions.
