Abstract     Background. Upon obtaining the written informed consent from the couple, the surplus embryos, resulting from in vitro fertilization procedure (IVF) at the Department of Obstetrics and Gynaecology Ljubljana, were frozen and stored. The frozen embryos of the patients who failed to conceive with fresh embryos, were thawed and transferred into the uterus. Methods. After pre-treatment of freezing solutions with succrose and cryoprotectant, embryos were stepwisely frozen to - 150 degrees C and stored in liquid nitrogen at - 196 degrees C. On the day of embryo transfer into the uterus (ET) embryos were thawed at room temperature in the series of thawing solutions. At most 3 survived embryos were transfeered into the uterus. For ovulation stimulation patients were pre-treated with human menopausal gonadotropins hMG (Pergonal, Serono) in doses which enabled developing as low number of preovulatory follicles as possible. Then, we waited for a natural ovulation, or it was induced by human chorionic gonadotropins (hCG). In patients with previous prooved ovulation cycles, the thawed embryos were transferred into the uterus in the spontaneous, non-stimulated cycle. Results. Until 1st February 1998 we froze 1410 embryos of 437 female patients, 3 embryos per patient on the average. The results of embryo thawing obtaine din the time period December 1996 - February 1998 were analysed. In this time period 110 embryo thawing cycles were performed in 110 patients. We thawed 302 embryos. 194 (64%) of embryos survived freezing and thawing processes.(Abstract truncated at 2000 characters)
Summary     Izhodišča. S postopkom zamrzovanja smo ob pisnem soglasju para shranili nadštevilne zarodke v postopku zunajtelesne oploditve (IVF) na Ginekološki kliniki v Ljubljani. Pri bolnicah, ki niso zanosile, smo zamrznjene zarodke odmrznili in jih vnesli v maternico. Metode. Zarodke smo po predpripravi v zamrzovalnih raztopinah s saharozo in krioprotektantom postopno zamrznili do - 150 degrees C in jih shranili v tekočem dušiku na - 196 degrees C. Na dan vnosa v maternico smo zamrznjene zarodke odmrznili na sobni temperaturi v zaporedju odmrzovalnih raztopin. V maternico smo vnesli največ 3 preživele zarodke. Za spodbujanje ovulacije so bolnice pred prenosom odmrznjenih zarodkov dobivale humani menopavzni gonadotropin hMG (Metrodin HP, Serono) v odmerkih, ki omogočajo razvoj čim manjšega števila predovulacijskih foliklov. Nato smo počakali na spontano ovulacijo ali pa smo jo izzvali z dodajanjem humanega horionskega gonadotropina (hCG). Pri bolnicah, ki so imele poprej dokazane ovulacijske cikluse, smo odmrznjene zarodke prenesli v spontanem, nespodbujenem ciklusu. Rezultati. Do 1. februarja 1998 smo zamrznili skupno 1410 zarodkov pri 437 bolnicah. Zamrznili smo povprečno 3 zarodke na bolnico. Analizirali smo rezultate odmrzovanja zarodkov v obdobju december 1996 - februar 1998. V tem obdobju smo opravili 110 odmrzovalnih ciklusov pri 110 bolnicah. Odmrznili smo 302 zarodka. Postopek zamrzovanja in odmrzovanja je preživelo 194 (64%) zarodkov. Zarodke smo presnesli v maternico v 91 (84%) odmrzovalnih ciklusov. Nosečnost smo dosegli pri 16 bolnicah, kar pomeni 17% stopnjo nosečnosti na prenos zarodkov v maternico (ET).(Izvleček prekinjen pri 2000 znakih)
