Abstract     Paratuberculosis is a chronic infectious disease caused by Mycobacterium avium subsp. paratuberculosis. To diagnose the disease the isolation and determination of the causing agent is necessary, which can take several months due to very slow growth of mycobacteria. We are introducing the method of polymerase chain reaction - PCR to detect the DNA of M.avium subsp. paratuberculosis which shortens the procedure to 2 to 3 days. In our study we have analysed the samples of intestines and mesenterial lymphatic glands obtained from eight serological positive ruminants, included wwere also positive and negative control and the control to determine the specificity of the reaction. Oligonucleotide primers P90 and P91 were used to amplify the nucleotide sequence of the insertion sequence IS900, characteristic for M.avium subsp. paratuberculosis. The products of PCR reaction were further amplified by nested PCR. The product of nested PCR was labelled and used as a probe in the dot blot hybridisation. The results of our study indicate that PCR could be used to detect M. avium subsp. paratuberculosis in the tissues of domestic animals.
