Abstract     Frankia is a filamentous nitrogen fixing bacterium. It is important as a symbiont on woody plants and forms root nodules on over two hundred dicotyledonous plants. Isolation and growing of Franikia in pure culture is difficult. In our research we have tried different methods of isolation. Root nodules of Alnus glutionosa and A. incana from different locations were used as sources of Frankia. Nodules were surface sterilized, crushed, filtered or ultracentrifugated and then inoculated into different liquid media. After incubation for over two months no growth was observed. Inoculation of alder seedlings with nodule crushates under laboratory conditions (in growing pouches) resulted in formation of nodules. Nodules were then checked for nitrogenase activity by acetylene reduction assay. Root nodules were also used for DNA extraction and molecular characterization of isolates based on amplification of ribosomal genes and genes for nitrogen fixation.
