Abstract     Doxicycline belongs to tetracyclines and is frequently used for therapy of Lyme borreliosis. We included doxicycline among antibiotics whose activity we wanted to test on borrelia. Experiments with this antibiotic were described before, but as in other cases, the authors used chemical methods for electron microscopy. Chemical reactions between fixative and biological sample lead to alterations of the latter and mask or interfere with the influence of the antibiotic.We decided to use cryomethods - hith pressure freezing combined with freeze substitution. In this way we prevented chemical reactions and were able to observe and study the action of antibiotic much more directly. Bacteria, grown on Modified kelly Pettenkofer (MKP) medium, were exposed to ceftriaxone at minimum inhibitory concentration (MIC) for 72 hours. Then we transferred bacteria to cel-lulose tubes, froze them by high pressure freezing method and dehydrated them by freeze substitution. Bacteria were embedded in Epon. Ultrathin sections were stained with uranyl acetate and lead citrate and observed in transmission electron microscope Jeol JEM 1200 EX II.
