Abstract     Several typing methods have been developed to explore the distributionand epidemiology of Clostridium difficile. One of commonly used is serotyping, which distinguishes the strains in more than 20 serogroups. Recently a typing scheme, based on differences in their toxin genes, was established, and variant strains were distributed to ten groups, toxinotypes. Variant strains are found only in some serogroups. We analysed strins of serogroups A1, A15 and E, which are most heterogenic as they contain strains similar to the reference strain, in addition to two or three different toxinotypes. To discover other genetic markers characteristic for variant strains, we applied pulsed-field gel elestrophoresis (PFGE) and AP-PCR to a selction of 28 strains. After PFGE of macrorestriction fragments of enzymes Smal and Nrul 13 and 15 different patterns were obtained, while AP-PCR with two different arbitrary primers gave 17 and 15 distinct patterns. After typing with PFGE and AP-PCR only epidemiologically related strains were grouped accordingly, which was especially with AP-PCR rarely the case for unrelated strains of the same toxinotype. Hence our results suggest that it is not likely that the toxinotypes are clonal subgroups of Clostridium difficile species.
