Abstract     The growth and development of two garlic cultivars Allium sativum L. cvs. Ptujski jesenski and Ptujski pomladanski was studied in tissue culture. The development of bulbs with different sucrose concentrations was also tested. B5 medium with 3% sucrose 5microM JA in 5microM 2iP was used for shoots was higher in cv. Ptujski pomladanski, on an avereage 13-15, than cv. Ptujski jesenski, on an avarage 12-13. After four weeks in culture, the basal plates were transferred to media without growth regulators but two different sucrose concentrations, 6 and 8%. Fourteen to eighteen bulbs formed on each basal plate after twelve weeks in culture. 8% sucrose had a stimulating effect on the bulb diameter in both cultivars. It was significantly higher in cv. Ptujski pomladanski than cv. Ptujski jesenski, regardless of the sucrose concentration in the media. Bulbs successfully germinated on the B5 medium with 3% sucrose and grew to maturity after transfer to in vivo conditions. The described procedure can be used for efficient micropagation of two seed garlic cultivars: Allium sativum L. cvs. Ptujski jesenski and Ptujski pomladanski.
