Abstract     Purpose: To analyse the expression of HLA-DR molecules on conjunctival epithelial cells by flow cytometry in glaucoma patients on long-term topical therapy. Methods: 7 glaucoma patients ( 8 eyes) and 10 controls (13 eyes) without ocular surface disease were included. The glaucoma patients were receiving two medications for at least one year. Collection of conjunctival epithelial cells was performed by imprint of ocular surface onto Millipore filter paper stripes after topical anaesthesia. Stripes of filter paper with adhered epithelial cells were put into phosphate buffered saline and incubated with monoclonal antibodies CD3, anti-cytokeratin and HLA-DR antibodies. Flow cytometric analysis was performed using a fluorescence activated cell sorter. Results: An average of 70 lymphocytes and 510 epithelial cells per conjunctival sampling was isolated. There was no statistically significant difference in the ratio of epithelial cells/lymphocytes between glaucoma and control patients. We found a significant increased expression of HLA-DR molecules on epithelial cells in glaucoma patients. The expression of HLA-DR molecules on T Lymphocytes in glaucoma patients was not significantly different from controls.
