Abstract     An ischemic in vivo model of urinary bladder was developed to study early molecular and ultrastructural changes of the mice urothelium. THe urothelium response to short-term experimental ischemia (30 to 120 minutes) was investigated by light, fluorescence and electron microscopy using immunocytochemistral and histochemistral methods as well as by flouremetric measurement of the (Ca2+)i. It was established that ischemic alterations are focally distributed and expressed by desquamation of all strata of the urothelium. Urothelium damages occur as funnels formed wounds extending to the basal cells. The damages are proportional to the duration of ischemia, contraction effect in vivo and in vivo induced reperfusion. Ischemia causes a disruption of the cell junctions and consequently the blood urine barrier. These changes are related to the increased (Ca2+)i and alternation of actin cytoskeleton. Exposal of desquamated live cells to the hypertonic and toxic urine gradually leads to irreversible changes.
