Abstract     Number of ribosomal RNA operons and sequence variability between 16S RNA gene copies in members of the rumen genus Prevotella were analysed. Chromosomal DNA was isolated, digested with seven restriction enzymes (BstEII, ScaI, SacI, Bg/II, Sall, XhoI and Hind/III), separated on agorose gel electrophoresis and transferred to nylon membrane. 16S rRNA and 23S rRNA genes were PCR amplifield, labelled with digoxigenin and used as hybridisation probes. According to number of the hybridisation signals P. albensis strains possess 4 or 5 rRNA operons, strains belonging to P. brevis possess 4 rRNA operons and P. bryantii strains possess 6 or 7 rRNA operons. Strains belonging to P. ruminicola probably possess 4 rRNA operons with one exception which possess 6 or 7 rRNA operons. Additionally, DNA fragments corresponding to the 16S rDNA hybridisation bands of P. ruminicola type strain were isolated from agarose gel and used as a template for PCR amplifications. Amplified 16S rRNA genes were used as templates for cycle sequencing of 425bp region on the beginning of genes. The sequences of the four genes differ at 6 to 48 positions. Differences between 16S RNA genes copies from rrnA, rrnB, rrnC, rrnD and ATCC 19189 (Accession. No. L16482) were 1,4% to 7,8%. 16S rRNA genes heterogeneities did not influence P. ruminicola phylogenetic position.
