Abstract     Introduction: In the central nervous system a neurotrophic compound nerve growth factor (NGF) is synthesised by neuronal and non-neuronal cells. Its synthesis and release can be modulated by certain neurotransmitters, cytokines, hormons etc. In the present work the influence of histamine on NGF release and molecular mechanisms, which regulate this process in primary cultures of astrocytes derived from newborn rats, was studied. Methods: Primary cultures of astrocytes were prepared from cortices of newborn rats. Histamine H1- and H2-receptor subtypes were identified and characterised by radioligand binding studies using 3H-mepyramine (Hl-receptor specific antagonist) and 3H-tiotidine (Hz-receptor specific antagonist), respectively. Enzyme-immuno assay (ELISA) was used to study the regulation of NGF release. Results: The co-existence of histamine receptor subtypes H1- and Hz- on astrocytes was confirmed with radioligand binding studies. There exists a single population of 3H-mepyramine binding sites with a equillibrium dissociation constant (KD ) of 3.5 nM and a maximal density of binding sites (Bmax) of 281 fmol/mg of protein. and a homogenous population of 3H-tiotidine binding sites with a KD of 1.8 nM and Bmax of 59 fmol/mg of protein. The displacement binding studies with different histaminergic ligands confirmed that 3H-mepyramine- and 3H-tiotidine-binding sites represent functional histamine H1- and H2-receptors, respectively. Astrocytes in primary cultures synthesise and release NGF. The basal levels of NGF release is 588.1+-67.6 pg NGF/mg of protein in 24 hours. The addition of histamine increases the NGF release from astrocytes to 1414.1+-59.7 pg/mg of protein, which is 2.3-fold of the control NGF secretion. (Abstract truncated at 2000 characters)
