Abstract     Five murine monoclonal antibodies (mAbs) were produced against B. burgdorferi strain pKo outer envelope proteins. Antigen specificities of mAbs were determined by Western blotting. MAb 1G7/A4/E8 was directed against the 31 kDa B. burgdorferi outer surface protein (Osp A), mAbs 1G6/G6/D12 and 2H7/H12/B1 against the 34 kDa B. burgdorferi outer surface protein (Osp B), while the remaining two antibodies recognized the 41 kDa protein, specific for the B. burgdorferi periplasmic cylinder axial filaments. All the mAbs were of IgG immunoglobulin class. Excellent activity in the indirect ELISA test and very low cross-reactivity to heterologous bacterial antigens indicated the high avidity and specificity of monoclonal antibodies for the B. burgdorferi antigens. The reactivity of mAbs in the indirect immunofluorescent test was also good, with an exception of 3B5/D12/A3 mAb. The fact that the flagellin specific 2A3/B9/H8 mAb was active in the indirect immunofluorescent test supports the thesis that certain flagellin epitopes might be exposed on the B. burgdorferi surface.
Summary     Pripravili smo pet monoklonskih protiteles, specifičnih za antigene zunanje ovojnice spirohete B. burgdorferi, sev pKo. Z imunsko-encimskim postopkom Western blotting smo dokazali, da je monoklonsko protitelo 1 G7/A4/E8 usmerjeno proti borelijskemu proteinu z molekulsko maso 31 kDa (Osp A), da monoklonski protitelesi 1G6/G6/D12 in 2H7/H12/B1 prepoznavata borelijski protein z molekulsko maso 34 kDa (Osp B), monoklonski protitelesi 3B5/D12/A3 in 2A3/B9/H8 pa protein borelijskih aksialnih filamentov z molekulsko maso 41 kDa (flagelin). Vseh pet monoklonskih protiteles pripada imunoglobulinskemu razredu IgG. Izrazita aktivnost v indirektnem testu ELISA govori o visoki afiniteti, zanemarljiva navzkrižna reaktivnost s heterolognimi bakterijskimi antigeni pa o veliki specifičnosti protiteles proti antigenom B. burgdorferi. Vsa pripravljena monoklonska protitelesa razen protiteles klona 3B5/D12/A3 so dobro reagirala tudi v indirektnem imunofluorescenčnem testu. Dejstvo, da je bilo v tem imunskem testu aktivno tudi monoklonsko protitelo 2A3/B9/H8, specifično za protein periplazemskih aksialnih filamentov, nakazuje možnost, da so določeni epitopi te beljakovine izpostavljeni na površini bakterijske celice B. burgdorferi.
