Abstract     Potato tuber necrotic ringspot disease, caused by necrotic strain of potato virus Y (PVY/NTN) is one of the most harmful potato diseases shown to have infected potato crops in Slovenia, cv. Igor being the worst affected. As the spreading of disease cannot be stopped by any of conventional control measures, we tried to obtain transgenic potato plants confering resistance to PVY/NTN. Two different coat protein (CP) gene constructs of PVY/NTN - sense or frame-shift were inserted into potato cv. Igor using Agrobacterium tumefaciens. Resistance of transgenic plants to virus PVY/NTN was evaluated by mechanical inoculation and by grafting-experiments. The symptoms on plants as well as on tubers were closely examined; the presence of PVY/NTN was tested by ELISA. Results showed resistance to PVY/NTN on three transformed lines from 19 tested. Resistance was identified by failure to develop symptoms on plants and on tubers after mechanical inoculation. One of these lines showed resistance also in grafting-experiments. New transgenic lines are under testing.
