Abstract     The present work deals with the study effects caused by the elevated body temperature in patients with fever on pharmacokinetics of three antibiotics: peniclin G, ciprofloxacin, and cefazolin. The mentioned physiological condition may namely affect the absorption, distribution and elimination of drugs. In the literature very few data about the influence of pharmacokinetics changes on the pharmacodynamics of antibiotics can be found, especially for the cases of human "in vivo" studies. Since pharmacokinetics of mentioned drugs fever is completely unknown the main objective of the study was to compare pharmacokinetics of three antibiotics during fever and defervescence and to evauate pharmacodynamic consequences of possible pharmacokinetic changes by developed compartment model and computer simulation using EAI 2000, analog-hybrid computer and MATLAB-SIMULINK simulation tool. The latter enables also prediction of drugs concentration profiles for higher dosage regimens used in severly ill patients. The main conclusion is that transition from febrile to afebrile stage changes the structure of pharmacokinetic model in most cases. In spite of that fact adjustment of the studied drugs dosage regimens during fever is not recommended.
