Abstract		Melanoma is an immunogenetic tumor with defined tumor antigens. An important therapeutic aim is the induction of anti-melanoma specific cytotoxic T-lymphocytes (CTL). Dendritic cells (DC) as natur's adjuvant have proven to be suitable for the inducation of anti-melanoma specific CTL in several murine models. We will discuss curent issues in the use of DC for treatment of human cancer such as the generation and application of human DC, use of surrogate helper antigens, various methods to detect antigen-specific immune response and briefly touch on some of our clinical data relating to vaccination of stage IV melanoma patients. Finally, we will propose reasons for missing response to DC therapy.
Summary		Melanom je imunogeni tumor z znanimi tumorskimi antigeni. Pomemben terapevtski cilj je indukcija specifičnih T limpocitov (CTL) proti melanomu. Dendritične celice (DC) so se v večjih mišjih modelih pokazale kot primerne za indukcjo specifičnih CTL proti melanomu. Razpravljali bomo o trenutnih rezultatih pri uporabi DC za zdravljenje raka pri ljudeh, kot je pridobivanje in uporaba humanih DC, uporaba nadomestnih helperskih antigenov, različnih metodah za odkrivanje za antigen specifičnih imunskih odzivov. Predstavili bomo tudi nekaj naših kliničnih ugotovitev, ki obravnavajo vakcinacijo bolnikov z melanomom v IV stadiju in razpravljli o vzrokih za neodzivnost na zdravljenje.
