Abstract     Receptor-mediated endocytosis occurs in all nucleated vertebrate cells and serves many important cellular functions. The trafficking of numerous membrane-bound receptors is mediated via clathrin-coated pits and is one of the best-characterised mechanisms of receptor-mediated endocytosis. The sequence of events leading to the entrance of G-protein coupled receptors (GPCRs) into the endocytotic pathway requiers a co-ordinated interaction between the receptor's intracellular domains and various proteins; both coated-vesicle components and intermediate connector molecules participating in endocytosis. The emphasis of this review is on clathrin-dependent receptor-mediated endocytosis, the intracellular pathways taken by internalized receptor/ligand, the methodology that can be applied to study receptor intracellular trafficking and the determinants regulating the efficiency of this process as exemplified by the gonadotropin-releasing hormone receptor (GnRG-R). The biological importance of GPCR internalization and the pathological processes associated with disturbances in endocytosis are also considered. The selected topics represent only a general insight into receptor-mediated endocytosis and do not cover all the current developments in this field.
Summary     Receptorsko pogojena endocitoza poteka v vseh jedrnih celicah vretenčarjev in je povezana s pomembnimi celičnimi funkcijami. Promet številnih membranskih receptorjev poteka po postopku pinocitoze z obloženimi mešički in je najbolje poznan mehanizem receptorsko pogojene endocitoze. Za niz dogodkov, ki privedejo do endocitoze s proteinom G združenih receptprjev (GPCR-jev), je potrebno usklajeno delovanje med znotrajceličnimi območji receptorjev in različnimi proteini, tako sestavinami mešička, obloženega s kaltrinom, kakor tudi "vmesnimi posredovalnimi molekulami" (adaptornimi proteini), ki sodelujejo v postopku endocitoze. V preglednem članku je opisana receptorsko pogojena endicitoza, znotrajcelični promet internalizirajočega receptorja/liganda in detreminante, ki vplivajo na učinkovitost tega procesa, kar je prikazano s primeri za receptor za gonadoliberin (GnRH). Prav tako so omenjeni biološki pomen internalizacije GPCR-jev in patološki procesi, ki sp posledica motenj v poteku endocitoze. Izbrana poglavja predstavljajo le splošen opis receptorsko pogojene endocitoze in ne vključujejo vseh novejših dosežkov na tem področju.
