Title     Correlation between liquid chromatography size-exclusion chromatography and Kjedahl method for determination of protein content in I. V. immunoglobulin fractions

Abstract     For thepharmaacopoeial distribution of molecular size of i.m. and i. v. Immunoglobulins size-exclusion chromatography (SEC) and liquid chromatography (HPLC)are available. Both methods are based upon the measurement of absorbance at A = 280nm. At this wavelength only the aromatic amino acids (tryptophan, tyrosine) exhibit the absorbance maximum. Therefore the protein content in fraction is largely determined by means of only two amino acids.Results have shown that on the basis of the absorption of aromatic amino acida at A = 280nm in the sample Immunoglobulin i.v. 3g(Biochemie, Vienna, Austria, batch no. 77487), the protein content of immunoglobulin fractions can be determined by both methods. This has been confirmed by a protein (nitrogen) percentage determination by the Kjeldahl method and the nitrogen percentage by C.H.N. elementary microanalysis in the dimer and the monomer immunoglobulin fraction of the same sample. The fractions were acquired by size-exclusion chromatography (SEC).
