Abstract     Hyperkinetic syndrome or attention-deficit hyperactivity disorder (ADHD) is the most frequent psychiatric disorder in childhood. It is a serious condition which can interfere with normal development of the affected child. Genetic factors are thought to be important components of ADHD etiology. Several associations between different candidate genes and ADHD were published so far. The best association is established for the DAT1 gene, which codes for a receptor for the psychostimulant drug methylphenidate. Mutations in coding regions of different candidate genes were not described so far. ADHD appears to be a complex trait, with many genes making small contributions to its etiology. Further research on genetics of ADHD will help to better understand this disorder and to improve the treatment of affected children.
Summary     Hiperkinetični sindrom je najpogostejša psihiatrična motnja v otroštvu. Predstavlja resno oviro za normalen razvoj otroka. Pri nastanku hiperkinetičnega sindroma igrajo pomembno vlogo različni genetski dejavniki. Opisane so bile statistično pomembne povezave med številnimi kandidatnimi geni in fenotipom hiperkinetičnega sindroma. Najbolje potrjena je povezava hiperkinetičnega sindroma in gena DAT1, čigar protein DAT1 je eno od prijemališč zdravila metilfenidata. Mutacije v kodirajočih delih različnih kandidatnih genov še niso bile opisane. Zato predpostavljajo, da je hiperkinetični sindrom posledica delovanja številnih genov, ki k nastanku sindroma posamično prispevajo zelo malo. Razjasnitev vloge genetike v hiperkinetičnem sindromu bo pomembno prispevala k razumevanju sindroma in izboljšanju obravnave prizadetih oseb.
