Abstract     Seminal experiments of Golub and coworkers demonstrated that tetracyclines inhibit matrix metalloproteinases and thus influence the breakdown of connective tissue and bone. This discovery led to the development of a formulation containing a subantimicrobial dose of doxycyclin (Periostat) and its use in patients with moderate and advanced periodontitis. The first multicentric study of Periostat therapy used as an adjunct to scaling and root planing showed significant improvement of clinical attachment and reduction of probing depth in treated patients. Further research and development of chemically modified tetracyclines with non-antibiotic properties will open new possibilities for the treatment of periodontal disease and other systemic disorders affecting the breakdown and restoration of connective tissue and bone.
