Abstract     Objective: To examine if ANCA-s of IgG and IgA isotype are present in serum patients with past history of Purpura Schoenlein-Henoch and persistent urinary abnormality findings and 15 healthy controls. Design: We retrospectively reviewed the medical records of patients with history of Purpura Schoenlein-Henoch between 1 January 1991 to 31 December 1994, and persistent renal abnormalities on follow-up (range 6 months - 40 months, mean 18 months) before a ANCA-test was done. All patients were assessed at the Department of Paediatrics Maribor Teaching Hospital. Methods: Within 48 hours after serum samples were obtained, they were tested for IgG and IgA ANCA by indirect immunofluorescence microscopy, a standard procedure described in detail by Allan Wiik 1989, and confirmed at the Third International Workshop in Washington 1990. The screening serum dilution was 1:20, (c-ANCA serum displayed a characteristic diffuse granular cytoplasmic staining pattern, p-ANCA a perinuclear staining pattern, atypical ANCA - with diffuse homogeneous cytoplasmic pattern. After immunofluorescence testing, sera were stored at -70 degree C, before tests were done for anti-MPO and anti-PR3 antibodies with use of commercialiy available enzyme-linked immunosorbent assays (ELISA), Euroimun (Luebeck, Germany). The assays were performed on aliqots of stored serum in accordance with the manufacturers procedural instructions. Results that exceded 20 RU (relative units) were considered positive for antibodies to MPO and PR3, respectively. Positive controls were sera from patients with Wegener's disease, negative controls were sera from 15 healthy children. Patients: 20 children (12 female, 8 male, age 4 - 15.2 years, mean 8.8 year.
