Abstract     Histochemical and cytochemical techniques were used to study the characteristics of the midgut during the development of honeybee, Apis mellifera, larvae. Free acid phosphatase appears in the cell cytoplasm before morphological signs of cell death during normal larval devlopment. Immunohistochemical analyses were made after p.o. and p.c. inoculation with Paenibacillus larvae var. larvae, causative agent of American foulbrood disease in honeybee larvae where extensive induced or necrotic cell death was observed. Hsp 70 was localised in Paenibacillus infected midgut epithelial cells and was not found in healthy larval tissue. In non infected larvae exposed nuclear histone protein, confirmed programmed cell death during normal tissue turnover, but in Paenibacillus infected larvae DAB reaction product was found distributed in all cell nuclei and also in cell cytoplasm. In necrotically deleted midgut cells pyronin staining was absent and dense red pyronin staining was found in nuclei and in the cytoplasm of non infected cells. The use of histo- and immunohistochemical techniques and their applications in honeybee pathology are also discussed in this review.
Summary     Histokemične in citokemične tehnike so bile uporabljene pri študiju značilnosti sprememb v srednjem črevesu pri razvoju ličinke medonosne čebele, Apis mellifera. Prosta kisla fosfataza se pojavlja v celični citoplazmi pred morfološkimi znamenji odmiranja celic pri fiziološkem razvoju čebelje ličinke. Imunohistokemične analize so bile narejene po p.o. in po p.c. inokulaciji Paenibacillus larvae var. larvae, povzročiteljem hude gnilobe čebelje zalege, ki povzroči obsežno inducirano ali nekrotično odmiranje celic. Stresni proteini (Hsp70) so bili ugotovljeni v srednjem črevesu okuženih ličink, v neokuženih ličinkah pa aktivnost Hsp70 ni bila ugotovljena. V neokuženih ličinkah je prisotnost histonskih proteinov v jedrih celic pokazatelj programiranega odmiranja celic pri fiziološki obnovi tkiv. V okuženih ličinkah s P. larvae pa je bila pozitivna rekacija DAB ugotovljena v jedrih vseh celic in tudi v njihovi citoplazmi. V nekrotično spremenjenih celicah srednjega črevesa ni bilo značilnega obarvanja s pironinom, značilno obarvanje pa je bilo ugotovljeno v jedrih in v citoplazmi neokuženih celic. V prispevku nadalje obravnavamo rabo histoin imunohistokemičnih metod v patologiji čebel.
