Abstract     The skeletal muscle of an adult rat is capable of adaptation to different functional demands. This ability differs from slow to the fast muscle. The limited adaptive range may be due to either different intrinsic properties of myogenic stem cells from which the muscle develops or to extrinsic factors. To test these assumptions we injured soleus (SOL) and extensor digitorum longus (EDL) muscles of the rat by bupivacaine immersion. We transplanted SOL or EDL to the EDL muscle bed, where muscles were allowed to regenerate innervated by the "fast" nerve n. peroneus. Six months later we analysed four regenerated SOL and four regenerated EDL muscles. Five rats of the same age as the experimental animals serve as control group. We proved myosin heavy chains (MHC-s) type 1, 2A, 2B and 2X/D in tissue slices by specific monoclonal antibodies using indirect imunoperoxidase method. We detected fibres expressing MHC-2X/D histochemically by adenosine triphosphatase (ATP-ase) reaction after preincubation in alkaline and acidic media. We determined the areal density of fibres expressing particular MHCs stereologically. We detected myosin heavy chain isoforms in muscle homogenates by SDS-gel electrophoresis and quantified their ratio densitometrically. (Abstract truncated at 2000 characters)
