Abstract     Background: The aim of this study was to establish the existence and the expression of the MAGE-3 gene in the breast's ductal invasive carcinoma (DIC) of different differentiations. The autor wanted to find out, which standard prognostic risk factor is in statisticaly significant correlation with expression of the MAGE-3 gene and how strong prognostic factor is the expression of MAGE-3 gene by itself in DIC of the breast. Because MAGE genes codes distinct antigens, which are recognised on various tumor cells by autologous cytolytic T lymphocytes, these antigens could constitute useful targets for specific immounotherapy. Methods: In this retrospective study 30 wel differentiated DICs, 51 moderate differentiated DICs and 63 poorly differentiated DICs were included. All specimens were before the imunohistochemistry reexamined and classified in concordance to standard prognostic factors: Immunohistochemistry with monoclonal antibodies against MAGE-3 and estrogen receptors were performed on the representative specimens of the DICs. Statistical methods: Studend T test, Spearman's correlation, Pearson Chi Square test, Multivariat analysis (Discriminant). Results: Out of 144 tumor tissue specimens of primary invasive ductal cancers, the expression of MAGE-3 gene was recognised in 48 (33,3 %) tumors, respectively. The expression of the MAGE-3 gene in DIC of the breast is in significant correlation with the tumor's differentation (p<0,0005), with expression/existence of the tumor necrosis (p<0,005) and with the invasion in lymph vessels (p<0,05). The correlation among the expression of the MAGE-3 gene and the expression of the estrogen receptors, diameter of the tumor, status/clearence of the axillary lymph nodes, peri- and intratumor inflammatory reaction, calcifications in the tumor, presence/amount of the tumor stroma and perineural invasion, statisticaly is nonsignificant (p>0,05).
