Abstract | 17beta-hydroxysteroid dehydrogenase (17beta-HSD) from the fungus Cochliobolus lunatus was purified to homogeneity and sequenced. The 810 bp open reading frame coding for a 270 amino acid protein covered all typical domains of the short-chain dehydrogenase/reductase protein family. Homology to several fungal carbonyl reductases as well as bacterial 7alpha-HSD and even mammalian 17beta-HSDs was determined. The results suggest that 17beta-HSD activity arose in lower cucaryotes and thus the steroid signalling system might have evolved very carly in evolution. | |
Summary | Iz glive Cochliobolus lunatus smo izolirali 17beta-hidroksisteroidno dehidrogenazo (17beta-HSD) in ji določili aminokislinsko in nukleolidno zaporedje. Kot smo ugolovili, zaporedji pokrivata vse značilne domene proteinov iz družine kratkoverižnih dehidrogenaz/reduktaz. Primerjava aminokislinskega zaporedja je pokazala, da je protein homologen nekaterim glivnim karbonilnim reduktazam, bakterijski 7alfa-HSD in celo sesalskim 17beta-HSD. Kot kažejo rezultati, se je 17beta-HSD aktivnost pojavila že pri nižjih evkariotih in se je verjetno tudi steroidni signalni sistem razvil že zelo zgodaj med evolucijo. | |
