Abstract     Aim: Direct contact of bacteria Borrelia burgdorferi sensu lato (B. burgdorferi s.l.) with a culture of endothelial cells (EC) induce EC expression of cell adhesion molecules (CAM), such as E-selectin and vascular cell adhesion molecule-1 (VCAM-1), but it was not known, whether different species of B. burgdorferi s.l. cause different intensity of E-selectin and VCAM-1 expression. Our aim was to test the hypothesis, that different species of B. burgdorferi s.l. can stimulate EC to different intensity of E-selectin and VCAM-1 expression. Method: We measured the differences in the expression of E-selectin and VCAM-1 in the culture of EC under the influence of different species of B. burgdorferi s.l. We used EC culture (HUVEC) and cultures of B. afzelii, B. burgdorferi s.s. and B. garinii. We added to a culture of EC different species of B. afzelii, B. burgdorferi s.s., B. garinii and incubated them at 37o C for 1 to 6 hours for the investigation of E-selectin and for 2 to 12 hours for the investigation of VCAM-1 expression. After the incubation immunohistochemical reactions with whole-cell ELISA were made on stimulated EC. The values of optical density (OD), which reflect the expression of CAM, were measured with a microplate reader. Results: Comparison of the expression of E-selectin and VCAM-1 on the EC culture under the influence of B. afzelii, B. burgdorferi s.l. and B. garinii revealed statistically significant differences (p<0,05). The intensity of the expression of E-selectin under the effect of B. afzelii was after 3 hours significantly higher and after 6 hours significantly lower than with the effect of B. burgdorferi s.s. or B. garinii. The intensity of the expression of VCAM-1 under the influence of B. afzelii was significantly lower in comparison with the B. burgdorferi s.s. and with the B. garinii. With the comparison of the OD values between B. burgdorferi s.s. and B. garinii statistically significant differences could not be demonstrated.
