Abstract     Exocytosis involves fusion of neurotransmitter- and hormone- containing vesicles with the plasma membrane. In neurones and neuroendocrine cells this process can be very fast and not easilly studied by standard biochemical methods. Here we describe an approach to monitor fast secretory activity in single cells by measuring membrane capacitance and intracellular Ca2+ dynamics using whole-cell patch-clamp technique and furaptra based Ca2+ photometry. As an example, the biochemical nature of regulated exocytosis in rat melanotrophs and cortical astrocytes is examined.
Summary     Med eksocitozo se zlijeta plazemska membrana in membrana sekretornega mešička, ki vsebuje hormon ali nevrotransmitor. V živčnih in nevroendokrinih celicah je ta proces tako hiter, da ga s standardnimi biokemijskimi metodami težko sledimo. Pričujoče delo opisuje poskuse, pri katerih smo prek meritev kapacitivnosti celične membrane pri visoki časovni ločljivosti zasledovali sekrecijsko aktivnost posamezne celice in hkrati fotometrično merili citosolno aktivnost kalcijevih ionov. Odvisnost hitrih procesov eksocitoze od kalcija smo proučevali pri dveh podganjih celičnih modelih, celicah srednjega režnja hipofize ter kortikalnih glia celicah.
