Abstract     Nicotiana tabacurn L. cv. Samsun tobacco plants were infected with two strains of Agrobacterium rhizogenes, ATCC 15834 and ATCC 11325, to transform plant cells and establish hairy root cultures. Strain ATCC 15834 was more efficient, and transformation succeeded on 100% of tobacco explants. The growth of the main root axis was also faster and more secondary roots formed compared with the culture established by strain ATCC 11325. Plant growth regulators are important endogenous and exogenous factors in the process of establishing of hairy root cultures. lAA and JA are known to stimulate the formation of secondary rools on non-transformed plants. Their effect on the growth of tobacco hairy root cultures was investigated using two strains of Agrobacterium rhizogenes. Added to the culture medium, JA and IAA affect both cultures similarly. JA in concentrations of 1-10 microM inhibited the growth of the main root axis, while IAA inhibited growth at concentrations of 0.1-10 microM. Lower concentrations (0.01 microM) of both hormones had no effect on the length of the main root axis. 10 microM JA and 10 microM IAA inhibited the number of secondary and tertiary roots. The inhibitory effects of high JA and IAA concentrations were also reflected in the fresh and dry weights of the cultures.
Summary     Tobak Nicotiana tabacum L. cv. Samsun smo okužili z dvemi sevi bakterije Agrobacterium rhizogenes ATCC 15834 in ATCC I 1325 ter tako vzpostavili laskaste koreninske kulture. Sev ATCC 15834 je bil bolj učinkovit. Infekcija izsečkov tobaka je bila 100%. Tudi dolžina glavne korenine in število stranskih korenin sta bila večja v primerjavi s kulturo vzpostavljeno s sevom ATCC 11325. Rastni regulatorji so pomemben endogen in eksogen dejavnik pri tvorbi laskastih koreninskih kultur. Indol ocetna kislina (IAA) in jasmonska kislina (JA) izboljšujeta rast stranskih korenin pri netransformiranih rastlinah. Zato smo preučevali učinek IAA in JA na rast laskaste koreninske kulture tobaka, vzpostavljeni z obema sevoma. Učinek JA in IAA, ki sta bili dodani gojišču je bil podoben pri obeh kulturah. 1-10 mikroM JA je zavirala rast glavne koreninske osi, IAA jo je zavirala pri koncentraciji 0.1-10 mikroM. Nižje koncentracije (0.01 mikroM) obeh uporabljenih hormonov niso imele učinka na dolžino glavne osi korenine. 10 mikroM JA in 10 mikroM IAA sta zaviralno delovala tudi na število sekundarnih in terciarnih korenin. Zaviralni učinek visokih koncentracij IAA in JA se je pokazal tudi pri meritvah suhe in sveže teže korenin.
