Abstract     Normal urinary bladder urothelium of adult mammals is stable epithelium with very slow turnover. It is three cell layers deep. Single intraperitoneal dose of cyclophosphamide causes necrosis, because of which urothelial cells are removed. Denudation is followed by regeneration, which is seen as hyperplasia, gradual differentiation of apical plasma membrane of the superficial cells and restoration of normal tree-layered urothelium. The purpose of this thesis was to establish whether apoptosis has a role in this process, in which layers it occurs, and differences in regeneration of upper, middle and lower part of the bladder. Apoptotic cells were detected by electron microscopy and by using TUNEL assay (light microscopy). It was shown that apoptosis plays a crucial role in regeneration. It appears in basal, intermediate and superficial layer of urothelium. Histological structure of regenerating urothelium and intensity of apoptosis are the same in the upper, middle and lower part of the urinary bladder. Our results show, that apoptotic cells are probably removed from the urothelium by two mechanisms: they are phagocytosed and digested by neighbouring urothelial cells or removed from the superficial layer into the lumen of urinary bladder.
