Abstract     In search of therapeutically useful muramyl dipeptide related analogues, a series of phthalimido desmuramyl dipeptides has been synthesized. Immunobiological properties of compounds were examined in an in vivo immunorestoration test. LK-413 and LK-423 were selected for further ex vivo immunological studies to investigate their ability to influence the IL-2 production and activation of macrophages. Jernes plaque forming cells assay was performed to monitor the effect of B cell differentiation. Blast transformation of T cells stimulated with ConA was used to detect the influence on T lymphocytes. The immunossupressive or immunostimulating activity of the compounds tested can be interpreted as a result of attachment of different parts of the molecule to different immunocompetent cells that show their specific responses in a dose dependent manner.
Summary     V iskanju terapevtsko uporabnih analogov muramil dipeptida smo sintetizirali serijo ftalimidnih dezmuramil dipeptidov. Imunološke lastnosti smo preizkusili v in vivo testu imunorestavracije. Spojini LK-413 in LK-423 sta bili na tej osnovi izbrani za nadaljnje ex vivo imunološke preiskave, da bi ugotovili njihov vpliv na produkcijo IL-2 in aktivacijo makrofagov. Jernov test blastne transformacije smo uporabili za spremljanje vpliva spojin na diferenciacijo celic B, vpliv na celice T pa smo vrednotili z blastno transformacijo limfocitov T po aktivaciji s ConA. Imunozaviralno ali imunospodbujevalno delovanje testiranih spojin lahko razložimo kot rezultat vezave različnih delov iste molekule na različne celice imunskega sistema. Jakost njihovega odziva je odvisna od odmerka testiranih spojin.
