Abstract     Floating tablets offer a new possibility of treating the stomach infected with Helicobacter pylori. The objective of this study was to select suitable materials forthe formulation of floating tablets with sustained drug release properties. In the preformulation studies, the differences between cellulose polymer (HEC, HPC, HPMC, K4M, Avicel) isotherms were estimated using, the DVS method and disappearance of the gas-generating agent at higher relative humidities was observed. The correlation between the wettability and the floating lag time proves that the first contract with water is not essential for good floating. Tablets with an incorporated drug stored at high relative humidity had a lower crushing force, the floating lag time increased, but the release profile did not change. Optimally designed tablets with ciprofloxacin hydrochloride monohydrate have good resistance to crushing, a floating lag time of less than 1 minute and float longer than 24 hours. The fact that they anable a more than 8-hour-long controlled drug release from non-distintegrated matrices plays an important role in prolonging the gastric residence time.
Summary     Plavajoče tablete predstavljajo novo možnost za zdravljenje infekcij želodca s Helicobacter pylori. Namen študije je bil izbrati primerne materiale za oblikovanje plavajočih tablet z zadržanim sproščanjem. V preformulacijskih študijah smo določili razlike med izotermami celuloznih polimerov (HEC, HPC, HPMC, K4M, Avice) z uporabo DVS metode in opazili izginevanje sredstva za tvorbo plina pri višjih relativnih vlagah. S korelacijo med močljivostjo in časom splavanja tablet smo dokazali, da prvi stik z vodo ni bistvenega pomena za dobre plovne lastnosti. Tabletam z vgrajeno učinkovino, shranjenim v vlažnem okolju, se zniža trdnost, podaljša se čas splavanja, profil sproščanja učinkovine pa se ne spremeni. Najoptimalnejše izdelane tablete s ciprofloksacin hidroklorid monohidratom imajo primerno trdnost, splavajo prej kot v eni minuti in plavajo več kot 24 ur. Omogočajo več kot 8-urno nadzorovano sproščanje učinkovine iz nerazgrajenih ogrodij, ki imajo pomembno vlogo pri podaljšanju časa zadrževanja farmacevtskih oblik v želodcu.
