Abstract     Since the beginning of its therapeutical use, it was demonstrated that some patients produce, when treated with interferon (IFN), antibodies able to neutralize its immunological activity. The incidence of antibody production in studies differs substantiallly. The question has been raised whether differences in assay techniques rather than differences in the molecules might account for the differences in occurence of antibodies. In this report we summarize the evaluations of various aspects of the immunogenicity of IFN-alpha using human cells in vitro and mice for testing the relative immunogenicity of IFNs in iri vivo animal models. Investigated were the distribution of genes for IFN-alpha2a, IFN-alphab, and IFN-alpha2c in various cell populations and the factors that influence the immune response to IFN-alpha in mice (frequency, routes of injections, immunogenicity of IFN aggregates). Based on the animal studies it is proposed that similar factors may likewise affect the immunogenicity of IFNs administered to patients.
Summary     Že od začetka terapevtske uporabe interferonov (IFN) je znano, da nekateri bolniki med zdravljenjem z IFN razvijejo protitelesa, ki lahko nevtralizirajo njegove biološke učinke. Poročila o pogostnosti pojavljanja protiteles se v različnih študijah močno razlikujejo. Postavlja se vprašanje, ali so vzrok za to razlike različne metode merjenja protiteles, ali razlike v molekulah različnih vrst IFN-alfa. V članku povzemamo ugotovitve vrednotenja imunogenosti različnih vrst IFN-a z uporabo človeških celic in vitro in vrednotenja relativne imunogenosti in vivo na miših. Proučevali so razmerja med nivoji izražanja različnih genov za IFN-alfa2a, IFN-alfa2b in IFN-alfa2c v različnih celičnih populacijah in faktorje, ki vplivajo na imunski odgovor na IFN-a pri miših (pogostnost in način injiciranj, imunogenost agregatov IFN). Na osnovi študij na živalih sklepamo, da podobni faktorji prav tako vplivajo na imunogenost IFN pri bolnikih.
