Title     Vpliv antioksidanta na morfološke in funkcijske spremembe arterijske stene pri osebah z zmerno hiperholesterolemijo

Abstract     Vitamin E as an antioxidant vitamin reduces the susceptibility of low-density lipoprotein (LDL) cholesterol to oxidation and may have antiatherosclerotic effects. We tested the hypothesis that long-term vitamin E supplementation might favourably afžects early functional and morphological changes in atherosclerotic process as well the laboratory characteristics in subjects with hypercholesterolemia. Forty - four asymptomatic men with untreated hypercholesterolemia and without other risk factors for atherosclerosis and any other systemic disease predisposing them to endothelial dysfunction or morphological changes on arterial wall were included to the prospective, double blind study. The subjects were randomly assigned to the group which took 400 mg vitamin E daily and to the group which took the placebo for six months. The diameter of brachial artery at rest, after reactive hyperemia (causing endothelium-dependent vasodilation) and after sublingual glyceryltrinitrate (causing endothelium-independent vasodilation) were determined by ultrasonographic method (B mode) before and after intervention period. The intima - media thickness (IMT) of the carotid bifurcation and 1 cm below bifurcation of the common carotid artery (CCA) was measured to follow early morphologic atherosclerotic changes. At the beginning of the study there were performed some clinical measurements (body weight, body height, body mass index, heart rate, blood pressure), laboratory parameters (hematological parameters, blood sugar, total cholesterol, LDL and HDL cholesterol, triglycerides, lipoprotein (a), values of autoantibody for oxidised LDL) as well coagulation and fibrinolytic characteristics (tissue plasminogen activator (t-PA) activity and antigen, tissue plasminogen activator inhibitor (PAI) activity and antigen, fibrinogen).
