Abstract     A number of studies have shown that inflamed periodontal tissues contain TNF-alpha, a proinflammatory cytokine which plays an important role in the development of periodontal disease. In this study, conducted on rats, the influence of recombinant human TNF-alpha (rhTNF-a) on inflammatory reaction and periodontal breakdown was analyzed in a model of experimental periodontitis, induced by placing silk ligatures around the maxillary right second molar. Ten rats were divided into two groups with five animals in each. The first group received rhTNF-alpha, applied subcutaneously via osmotic pumps over a period of 2 wk; in the second group, PBS was administered in the same manner. Inflammatory reaction and degradation of periodontal tissues were evaluated morphometrically on HE stained sections. When used alone, ligation resulted in accelerated periodontal degradation, while rhTNF-alpha had no significant effect. The combined use of rhTNF-alpha and ligation resulted in a significantly greater inflammatory reaction and periodontal breakdown than either treatment alone. We conclude that subcutaneous administration of rhTNF-alpha accelerates the progression of experimental periodontitis in rats.
