Title     Comparison of polar lipid profiles of Clostridium difficile isolates from different geografical locations

Abstract     Clostridium difficile is a known pathogen of antibiotic-associated diarrhoea (AAD) and pseudomembranous colitis (PMC) and has been isolated from numerous hospital and community outbreaks. The aim of this study was to test the hypothesis that geographical differences occur in strains of C. difficile of British, Belgian, and French origin with respect to their phospholipid analogue profiles. Lipids were extracted with chloroform-methanol and examined by fast atom bombardment mass spectrometry in negative-ion mode. The major higher mass peak for all Manchester isolates was m/z 747 attributable to the expected presence of PG (34:1) while other isolates showed different major peaks of m/z 703, 677, 747, 733 and 705 with putative identities PG (31:2); PG (29:1); PG (34:1); PG (33:1); PG (31:1). Phospholipid analogue distributions of all pairs of isolates were compared, using the Pearson coefficient of linear correlation (r). These coefficients were used to produce a dendrogram by single-linkage clustering. Isolates from Manchester, U.K., clustered separately from other isolates. A single strain, from Birmingham, U.K. clustered with strains from Belgium and France. Thus, C. difficile shows both quantitative and qualitative differences in polar lipid profiles that may correlate with geographical origin.
