Abstract     The maintenance of tissue architecture depends upon the controlled interaction between epithelial and mesenchymal cells. Nevertheless, the role of lamina propria in urothelial proliferation and differentiation in vivo and in vitro is still poorly understood. Therefore, the aim of this work was to elucidate the effect of lamina propria on the morphology and differentiation of urothelial cells in vitro. We studied regulation of regeneration of explant wounded area. In our early studies we showed that explant outgrowths consisted of well differentiated urothelial cells. Now we studied the origin of the differentiated cells in primary urothelial cell outgrowths. Four different primary explant cultures of mouse urinary bladder were established; cultures of type I and II were grown with urothelium and lamina propria, cultures type III with the explant of urothelium and cultures type IV were grown with the explant made only of lamina propria. In cultures type II the explants were removed on the 2., 4., 7., 10 and 14. day of culturing. We did not succeeded establish the secondary urothelial cell culture from the cultures type I. Cultures were maintained on Cyclopore membranes in serum free medium for 2, 4, 7, 10, 14 and 21 days. We used fluorescent and confocal light microscopy and transmission and scanning electron microscopy to analyse the morphology of urothelial cells and the presence of antigenic and ultrastructural markers associated with urothelial cytodifferentiation. The epithelialization of cut edges of the explant began within 24 hours and by 48 hours exposed lamina propria was covered by a two-layered, rarely by a three- or four-layered urothelium with the terminally differentiated urothelial cells on the surface.
