Abstract     Background Neuromuscular junction and the enzyme acetylcholinesterase (AChE) have been studied for decades. Until now researchers have studied the impact of altered pattern of muscular activation, denervation and elevated serum concentration of thyroid hormones on the expression of muscle AChE. Aim With this study we examined the expression of AChE in the fast muscle extensor digitorum longus (EDL) and the slow soleus muscle under conditions of hypothyroidism and reinnervation of muscle. Hypothesis We assumed that the levels of AChE mRNA of hypothyroid rats in fast and slow muscles falls. Reinnervation of the soleus muscle causes high rise of AChE mRNA above its normal value and in the EDL muscle a slow rise towards normal value. We also assumed that hypothyroidism acts against the effects of reinnervation on the level of muscle AChE mRNA. Methods Adult male rats were used. We investigated the following groups: group of rats made hypothyroid for 4 and 8 weeks, group of rats with reinnervated soleus and EDL muscles 21 days after n. ishiadicus crush, and the group of hypothyroid rats with reinnervated soleus and EDL muscles. Levels of AChE mRNA in muscles were determinated by the Northern blot analysis. The levels were expressed as the percentage of the control level in the EDL muscles. Data were statistically analysed. Results 4 weeks after removal of the thyroid gland the concentration of serum T3 fell to approximately 10% of normal. Level of AChE mRNA in the control soleus muscle was approximately 30% of normal EDL. After 4 weeks of hypothyroidism it fell statistically significantly to approximately 20%of the normal EDL.
