Abstract | Aims/Hypothesis: The cellular mechanisms for the insulin resistance in pregnancy and gestational diabetes mellitus are unknown. The plasma cell membrane glycoprotein PC-1 has been identified as an inhibitor of insulin receptor tyrosine kinase activity and may play a role in insulin resistance. The aim of the study was to examine the effects of insulin on glucose transport and changes in insulin receptor tyrosine phosphorylation, IRS-1 phosphorylation and PI-3 kinase activity in adipocytes from women with gestational diabetes mellitus. It was our objective to study a correlation between PC-1 content and tyrosine phosphorylation of the insulin receptor on one hand and PI -3 kinase activity on the other hand. In addition, a correlation between PC-1 content and adipocyte glucose transport was explored. We wanted to check the hypothesis that PC-1 content is associated with alterations in insulin signalling pathway in adipocytes from women with gestational diabetes mellitus. Methods Adipocytes were obtained either during elective caesarean section from three groups of subjects: Type 2 diabetic pregnant women (n=6) women with GDM (n=10) and pregnant women with normal glucose tolerance (n=6) as pregnant control subjects or during elective gynaecological surgery from nonpregnant (n=6) control subjects. Glucose uptake in adipocytes was measured radiometricaly using [14C-U] glucose. Basic and insulin-stimulated glucose transport was measured by using 14C labelled glucose, which was detected by radiometry. Effect of insulin on tyrosine phosphorylation of insulin receptor was determined by using immunoprecipitation and Western Blot techniques. (Abstract truncated at 2000 characters). | |
