Abstract     Epilepsy is a neurological disease arising from strong and uncontrollable electrical firings of a group of neurons in the central nervous system. Experimental epilepsy models have been developed in order to understand the physiopathology of the epileptic seizures. The aim of this study was to estimate the numbers of neurons in the hippocampus of penicillin induced epileptic rats, via a new stereological method "the optical fractionator". In experimental group, S00 IU penicillin-G was injected intracortically, and in control group, same amount of saline was administered. After one week, animals were decapitated and their brains were removed by craniotomy. Frozen brains were cut at a thickness of 150 `m in a cryostat. Sections collected via systematic random sampling were stained with hematoxylen-eosin. Microscopic images of pyramidal cell layers from CA1,CA2,CA3 subdivisions of the hippocampus were then transferred to a monitor, using x100 objective. Neuronal counts were done by using the optical disector method in the frames determined with fractinator sampling scheme. The total pyramidal neuron numbers were estimated using the optical fractionator method. The total pyramidal neuron numbers were statisticaly lesser in the experimental group (mean = 142 888 ± 11 745) than the ones in the control group (mean = 177 953 ± 10 907) (p < 0.05). These results suggest that, decrease in the hippocampal neuronal numbers in penicilin model of epilepsy can be detennined objectively and efficiently by using the optical fractionator method.
