Abstract     The present work was based on the results of "in vivo" experiments on rats that had shown that hypercalcemia had lead to morphological and biochemical hyperfunction of follicular cells. Regulation of activity of follicular cells should directly, or indirectly via paracrine action of serotonin secreted from parafollicular cells, depend on presence of calcium ions. Effect of calcium, serotonin and their combination, in presence and absence of thyrotropin, was studied on a cell line of rat follicular cells by using two methods: measuring of quantity of the produced cAMP (cyclic adenosine 3', 5'-monophosphate) and measuring of 3H-timidin incorporation into cell DNA. Results show that serotonin does not stimulate the cells while at high concentrations (10 mM) it has toxic effects. At high concentrations (30 mM) calcium stimulates follicular celis, however, not via cAMP. It probably acts via the phosphatidylinositol - protein kinase C pathway. As expected, thyrotropin stimulates follicular cells; at higher concentrations (3mM, 10mM, 30mM) calcium diminishes its effects, probably by activation of a cAMP phosphodiesterase wich disintegrates cAMP.
