Abstract | Several methods are available for identifying and typing M. avium complex strains for taxonomic or epidemiological purposes. These methods are either. phenotypic or genotypic. Serotyping, once the most widely used phenotypic method, allows the classification of M. avium complex strains into serovars 1 to 28 and the three additional serovars - 41 to 43 that represent M. scrofulaceum. Other phenotypic methods include multilocus enzyme electrophoresis, phage typing and several types of chromatography. Recently genotypic methods such as pulsed-field gel electrophoresis (PFGE), typing with plasmids, polymerase chain reaction (PCR), sequencing, spoligotyping and restriction fragment length polymorphism (RFLP) analysis have replaced the phenotypic methods. By using genotypic methods it is possible to confirm relationships discovered with standard methods or to reveal new ones. Seven different RFLP patterns of M. avium complex strains in Slovenia were identified using IS901-RFLP analysis. | |
Summary | Za identifikacijo in tipizacijo sevov mikobakterij sklopa Mycobacterium avium v taksonomske ali epidemiološke namene je na voljo več metod, tako fenotipskih kot genotipskih. Med fenotipskimi se je nekdaj največ uporabljala serološka tipizacija, na osnovi katere je sklop M. avium razdeljen na 28 serovarov ter na tri dodatne serovare - 41, 42 in 43, ki predstavljajo M. scrofulaceum. Fenotipsko lahko omenjene mikobakterije tipiziramo tudi z multilokusno encimsko elektroforezo, različnimi vrstami kromatografij in s fagotipizacijo. Fenotipske metode so v zadnjem času nadomestile genotipske, kot so pulzna elektroforeza, tipizacija s plazmidi, polimerazna verižna reakcija (PCR), sekveniranje, spoligotipizacija in metoda določanja polimorfizma dolžin restrikcijskih fragmentov (analiza RFLP). Na podlagi le-teh lahko dokazujemo povezave, ki smo jih odkrili že s klasičnimi metodami, ali pa na novo odkrivamo take, ki jih s klasičnimi metodami nismo mogli ugotoviti. Z analizo RFLP na osnovi IS901 je bilo v Sloveniji ugotovljenih 7 različnih RFLP tipov mikobakterij sklopa M. avium. | |
