Title     Column separation of motile spermatozoa and its influence on the quality of equine semen

Abstract     Fifty ejaculates from six Lippizaner stallions were included in this study. Each ejaculate was divided into two parts. One part was processed using standard methods (SP) and the other part was processed using a column separation (CS) procedure to select spermatozoa of higher quality. Column separation significantly increased (P < 0.001) both motility (83.9 +- 0.7 % vs. 66.8 +- 1.3 %) and progressive motility (72.4 +1.0 % vs. 54.8 +- 1.4 %) and a relative high recovery rate of motile spermatozoa (71.3 +- 1.1 %) was established. These results not only confirmed those from previous studies but also showed that CS was able to positively alter the results of freezing. Additionally, morphological studies were performed and these showed that in column-separated samples there was a significant decrease (P < 0.001) of spermatozoa with morphological abnormalities (31.5 +- 1.1 % vs. 54.3 +- 1.1 %). Column separation lowered the percentage of acrosome-damaged spermatozoa (7.6 +0.4 % vs. 22.4 +- 0.9 %) and sperm cells with protoplasmatic droplets (0.8 +- 0.1 % vs. 13.6 +- 0.8 %) but increased those with a coiled tail (17.7 +- 1.1 % vs. 7.8 +- 0.6 %). These results show that the column separation of stallion spermatozoa is an efficient method for quickly accumulating intact spermatozoa.
Summary     V raziskavo smo vključili 50 ejakulatov šestih žrebcev pasme lipicanec. Vsak ejakulat je bil razdeljen na dva dela. Prvega smo obdelali po standardnem postopku (SP), drugega pa po postopku kolonske separacije (KS). V vzorcih, ki smo jih obdelali po postopku KS, smo ugotovili statistično značilen (P < 0,001) dvig odstotka gibljivih (83,9 +- 0,7 % proti 66,8 +- 1,3 %) in progresivno gibljivih semenčic (72,4 +1,0 % proti 54,8 +- 1,4 %) ter relativno visok odstotek separiranih gibljivih semenčic (71,3 +- 1,1 %). S tem smo potrdili rezultate poprejšnjih raziskav, poleg tega pa nismo ugotovili negativnega vpliva prilagoditve KS postopku zamrzovanja. V raziskavo smo vključili tudi morfološko preiskavo vzorcev. Ugotovili smo statističo značilno (P < 0.001) znižanje odstotka morfološko spremenjenih semenčic (31,5 +- 1,1 % proti 54,3 +- 1,1 %). Največji vpliv KS smo ugotovili pri poškodbah akrosoma, v prisotnosti protoplazemskih kapljic in zavitih repov. Ugotovili smo povečanje odstotka zavitih repov (17,7 +- 1,1 % proti 7,8 +- 0;6 %) ter znižanje odstotka akrosomalnih poškodb (7,6 +- 0,4 % proti 22,4 +- 0,9 %) in prisotnosti protoplazemskih kapljic (0,8 +- 0,1 % proti 13,6 +- 0,8 %). Rezultati naše raziskave dokazujejo, da postopek KS omogoča hitro in uspešno prečiščevanje celotnega žrebčevega ejakulata.
